Place mother material in green house for atleast 4 weeks at 25-28C and wait fot the new sprouts. Use only upper 3-5 buds only as explant to raise in vitro shoots. Never used the mature shoots. washing as per lab norms vary from lab to lab. Medium: Simple 1/2 strehght ms medium with additional MgSO4.7H2O (170 mg) and NaH2PO4(170mg), sucrose (20 gm), pH (5.0) yeild fair results. The hormone balace is 2-ip@0.75 mg, IAA 0.05 mg) The first shoot with new leaf appear after three weeks and by excising basal part transfer to fresh medium and wait for juvenile crop once it established then adjust some hormone balance and go commercial.
